MACSima

Imaging Platform

The platform, by Miltenyi Biotec is a fully automated spatial biology system designed to map hundreds of protein and RNA biomarkers on a single tissue section at subcellular resolution.

The platform automates a continuous loop of staining, high-resolution imaging, and gentle signal erasure. By utilizing photobleaching or specialized release mechanisms instead of harsh chemicals, the system erases fluorescent signals without damaging the delicate tissue morphology. This gentle approach allows a single sample—whether it is FFPE, frozen tissue, or an organoid—to undergo dozens of consecutive cycles, enabling true sequential multiomics by analyzing both spatial transcriptomics and proteomics on the exact same slide.

To process the massive amounts of imaging data generated by these experiments, the platform integrates with MACS iQ View software. This specialized software translates complex image pixels into actionable biological insights through automated cell segmentation, phenotypic clustering, and deep spatial microenvironment analysis.

Services we provide:

The MACSima infrastructure operates as a joint initiative between StemTherapy (ST) and Lund University Bioimaging Centre (LBIC), combining technological access with advanced analysis expertise.

Hosting and Infrastructure:
• The MACSima instrument is hosted by StemTherapy and placed on BMC A11
• The analysis license is hosted by LBIC placed on BMC D11

The platform will operate primarily through:
• Supported access (ST)
• Standardized acquisition workflows (ST)
• Support for data analysis (LBIC)

Booking:

Contact: bengt [dot] mattsson [at] med [dot] lu [dot] se (Bengt Mattsson) BMC A1155, responsible for the instrument, user support, and coordination of acquisition workflows.

Supported access (ST)
• First contact envolved with people from MILTENYI will assist in consultation during experimental design and panel selection and guidance on sample preparation and acquisition parameters. Following the included user training, Miltenyi Biotec can offer regular sessions for users and core facility staff to come together and discuss experiment setup and analysis (free of charge).

Standardized acquisition workflows (ST)
• To book a acquisition session, you will contact bengt [dot] mattsson [at] med [dot] lu [dot] se (Bengt Mattsson) directly to coordinate the date, time, and specific arrangements.

Data Analysis Support (LBIC)
• LBIC will provide access to the MACSiQ View analysis software to all users through one of its local workstations at D11. Access and booking of the analysis station will be done through the LBIC booking system (InfinityX). New users will get basic training on the MACSiQ View software by LBIC staff. Users can receive advanced analysis support from Miltenyi Biotec.

Charges:

Each user covers all cost for consumables and antibody panels.
Users are responsible for data transfer from MACSima instrument to analysis workstation.
Users are responsible for data storage of own data after analysis is completed.

Cost for analysis:
Using the MACSiQ View software (including workstation usage): 100 SEK/hour (fixed for the duration of this agreement).
Basic training by staff: 550 SEK/hour (based on LBICs user fee model and subject to change).
